871 Higher-order chromatin organization heterogeneity among epidermal cells during development

Our study aims at explaining why the 3D higher-order chromatin structure developmentally controlled keratinocyte-specific loci of sub-population epidermal basal layer cells is organized in fundamentally different way. We performed analysis widefield and SIM super-resolution images mouse cryosections with FISH immunofluorescence. One still not fully understood aspects differentiation importance changes for epidermis-specific gene regulation. Big part genes involved are clustered several loci. This clustering important establishment their proper spatio-temporal expression pattern. Previous work showed that one those loci, complex (EDC), harboring almost 70 genes, relocates during development from nuclear periphery to interior. According results number decreases 70% E11,5 25% E16,5. change remarkable, however, it also raises questions about identity which distribution doesn't change. data show minority EDC localizing intra-nuclearly E11.5 or peripherally E16.5 belong groups: cycling G1 G2 phases keratinocytes. Careful shows distance between particular parts locus differ all cell types, central non-cycling keratinocytes more compact. explain we could observe patterns organization murine epidermis provides missing characteristic non-keratinocyte other sub-populations.
